Synthetic Phosphoethanolamine and the Analogy with Castor Oil-Based Biomaterials: A Hypothetical Extension of Chierice’s Research The synthetic phosphoethanolamine developed by Prof. Dr. Gilberto Orivaldo Chierice has been proposed as an innovative approach in oncology. Its mode of action has been described as a molecular marker for neoplastic cells that exhibit predominantly anaerobic metabolism, consistent with the Warburg effect, whereby tumor cells prioritize glycolysis even in the presence of oxygen, leading to excessive glucose consumption and lactate production. Chemically synthesized through an alternative route, synthetic phosphoethanolamine structurally mimics endogenous precursors of phospholipids such as phosphatidylethanolamine and phosphatidylcholine, which are crucial for the integrity of both cellular and mitochondrial membranes. The hypothesized mechanism of action involves signaling the immune system to induce selective apoptosis in malignant cells, mediated through modulation of mitochondrial potential and lipid dissolution of tumor cell membranes, without affecting normal cells. A less widely discussed hypothesis, here referred to as the “secret mechanism,” involves the hepatic biotransformation of synthetic phosphoethanolamine. In this model, the compound conjugates with fatty acids, forming a specialized phospholipid capable of infiltrating the membranes of anaerobic cells. This process would theoretically counteract the Warburg effect by restoring oxidative aerobic metabolism, promoting carbon–carbon bond cleavage into CO₂, and enabling tissue regeneration within a period of 6–8 months. This hypothetical framework can be logically compared with other biomaterials also developed by Chierice, particularly castor oil–derived bone prostheses (polyurethane scaffolds from Ricinus communis). These biomaterials are injected in a viscous state, solidify upon contact with blood, and function as biodegradable scaffolds that are progressively replaced by native bone tissue without eliciting immune rejection. From this perspective, synthetic phosphoethanolamine could be understood as a “molecular scaffold” in oncological applications, potentially guiding the transformation of tumor tissue into normal tissue. Such a mechanism, if confirmed, would represent a low-cost and accessible therapeutic approach, while also challenging conventional pharmaceutical paradigms. It must be emphasized, however, that the notion of a “hepatic phospholipid” capable of reversing the Warburg effect remains speculative and has not been experimentally demonstrated. This conceptual extension arises from an analogy with the castor oil–derived scaffolds, which themselves function effectively in practice despite incomplete mechanistic elucidation. In summary, synthetic phosphoethanolamine—hypothetically modified in the liver through conjugation with fatty acids—might form a novel phospholipid species capable of resetting the Warburg effect. This would, in theory, force tumor cells into oxidative respiration, trigger selective self-destruction, and promote tissue regeneration, in a manner conceptually parallel to the regenerative capacity observed in castor oil–based prosthetic scaffolds.
